I would have focused a lot on the conflict between the Thumb and Asher. In the few days that Sophie's soul is gone, Asher would obliterate the remaining Kurokumo clansmen. Afterwards... I don't know. Boris and Kalo would definitely be big antagonists for a long time, and Salvador/Dawn Office would play a big role in the conflict.
