I don't think it's anything specific, like an attack or spell, but just an all-around cumulative upgrade. More raw power, more flexibility, more stamina, etc. with using their seed and its abilities. Slyas's rune mastery and Will use takes a notable jump every time he's formed and assimilated a Virtue seed
By now, though, the Glutton was probably very close to becoming a True Gluttony Disciple, erasing the path for everyone that came after him. And as someone who was following the True Path, unlike the Rat and the Boar, he would be able to display real power as a Madness Disciple.
