Good theory but no. Remember Starswirl the bearded? greatest magician and master of both princess but he didn't become an alicorn. And according to the comics, candace became an alicorn for accident and so it did twilight, for what i know, there's no magic that allow a pony to become an alicorn.
The rune allows the object in question to be affected by the spell inscribed on it, absorbing the magic around us in order to activate it.
